Inhibiting PARP for pathologic vascular calcification

PARP inhibition could treat pathologic vascular calcifications. In patients with carotid artery disease, sites of calcification within atherosclerotic plaques and blood vessel tunica media had higher extranuclear deposition of PARP-synthesized poly(ADP-ribose) than non-calcified areas in diseased or normal arteries. In human vascular smooth muscle cells cultured under calcifying conditions, six PARP inhibitors, including Minocin minocycline, reduced calcification.
