PcrV-neutralizing mAbs for Pseudomonas aeruginosa infections
Patient-derived mAbs targeting PcrV, a pentameric structural protein that forms the needle-tip complex of the Pseudomonas aeruginosa type III secretion system, could help treat infections caused by this pathogen by preventing bacteria-induced host cell death.
In human lung cell lines, twenty mAbs derived from patients with cystic fibrosis chronically infected with P. aeruginosa inhibited host cell death with IC50s ranging from 0.010-19.72 µg/mL, and had greater inhibitory activity than the bispecific PcrV-Psl antibody gremubamab, which had previously failed in clinical trials to prevent pneumonia caused by P. aeruginosa. ...
