The American Heart Association's Stroke Council published guidelines in Circulation for the management of patients with acute ischemic stroke, recommending that patients be given tPA within three hours of stroke if they meet the criteria of the National Institute of Neurological Disorders and Stroke r-TPA Stroke (NINDS) Study.
In the NINDS study, which GNE used to support its NDA for Activase, patients were treated within three hours of stroke onset, with many treated within 90 minutes (see BioCentury Extra, June 7). The treatment group was given 0.9 mg/kg of tPA up to a maximum of 90 mg. There were significant improvements in the treatment group at 24 hours and at three months. While the rate of bleeding was higher in the tPA group, the rate of death or severe disability was less in the treated patients. ...
