Inhibiting S1PR1 for neuropathic pain

Rat and mouse studies suggest S1PR1 inhibitors or modulators could help treat neuropathic pain. In a rat model of neuropathic pain, astrocyte-specific knockout of S1PR1 in the CNS or intrathecal delivery of an siRNA against S1PR1 decreased pain-associated behaviors compared with a normal S1PR1 expression or a non-targeting control siRNA. In mouse and rat models of neuropathic pain, oral or intrathecal administration of S1PR1 antagonists or modulators, including Mayzent siponimod and ponesimod, decreased pain-associated behaviors compared with pretreatment baselines or vehicle.
