In vitro, mouse and rat studies identified a benzenesulfonamide-based CAII inhibitor that could help treat epilepsy. Chemical synthesis of benzenesulfonamide analogs and in vitro activity assays yielded a compound that inhibited CAII with a Ki of 33.2 nM. In mouse and rat models of epilepsy, the compound prevented seizures in five of six mice and five of six rats. Next steps could include testing the compound in additional models of epilepsy...
