Patient sample, cell culture and mouse studies suggest inhibiting GDF15 could help treat pancreatic ductal adenocarcinoma (PDAC). In patients, plasma and tumor levels of GDF15 were higher than in samples from healthy volunteers, and in the patients, high tumor levels of GDF15 were associated with poor survival. In a mouse PDAC cell line co-cultured with mouse peritoneal macrophages, knockdown of GDF15 in cancer cells decreased cancer cell survival compared with normal GDF15 expression. In two mouse models of PDAC, tumor-specific knockdown of GDF15 decreased tumor growth and tumor cell proliferation and increased tumor cell apoptosis, and in one of the models knockdown of GDF15 increased survival. Next steps could include identifying and testing GDF15 inhibitors in PDAC models...
