Mouse studies suggest topical tacrolimus could help treat lymphedema. In a mouse tail model of lymphedema, a topical formulation of tacrolimus decreased tail swelling and volume, soft tissue thickness, infiltration of inflammatory cells in the tail and markers of fibrosis, and increased markers of lymphatic function compared with topical vehicle. In another mouse model of lymphedema, the topical formulation of tacrolimus decreased dermal backflow and infiltration of inflammatory cells at the injury site and increased formation of lymphatic vessels.
