Therapeutics: Factor XIa

In vitro, mouse and rabbit studies suggest an anti-Factor XIa mAb could help treat thrombosis. Screening of an antibody library and mutagenesis of hits identified a mAb that inhibited human Factor XIa activity in vitro with an IC50 of 0.22 nM. In a mouse model of carotid artery occlusion transfused with the human Factor XIa precursor Factor XI, the mAb increased blood flow compared with a control antibody. In a rabbit model of venous thrombosis, the mAb delayed clotting and decreased clot weight. Next steps could include testing the mAb in animal models of other types of thrombosis...
