Therapeutics: Peroxisome proliferation activated receptor γ coactivator 1-α (PPARGC1A; PGC-1α)

Cell culture and mouse studies suggest promoting PPARGC1A expression could help treat breast cancer. In human breast cancer cell lines, a tool compound that promoted PPARGC1A expression decreased proliferation compared with vehicle. In a mouse mammary cancer cell line, PPARGC1A overexpression increased sensitivity to the antifolate drug methotrexate compared with normal PPARGC1A expression. In a mouse model of mammary cancer, PPARGC1A overexpression increased tumor sensitivity to methotrexate.
