Therapeutics: Monoacylglycerol lipase (MAGL)

Mouse studies suggest combining MAGL inhibitors with morphine could help treat neuropathic pain while limiting the development of morphine tolerance. In a mouse model of injury-induced neuropathic pain, co-injection of morphine plus a MAGL inhibitor tool compound decreased hyperalgesia, thermal allodynia and mechanical allodynia compared with morphine alone. In the pain model receiving daily injections of morphine for six days, daily injections of the MAGL inhibitor increased morphine's antinociceptive effect compared with vehicle. Next steps could include testing MAGL inhibitors in models of pre-existing morphine tolerance. ...
