Mouse studies suggest inhibiting tumor-associated macrophages (TAMs) with liposomal clodronate could help treat lung cancer. In a mouse model of Lewis lung carcinoma, a liposomal preparation of the bisphosphonate clodronate, which induces TAM apoptosis, plus the oxidative phosphorylation inhibitor metformin decreased tumor volume compared with vehicle plus metformin. Also in the model, treatment with liposomoal clodronate followed by ionizing radiation decreased tumor volume compared with vehicle plus radiation, and liposomal clodronate plus an anti-PD-L1 mAb decreased tumor growth compared with vehicle plus the anti-PD-L1 mAb. Next steps could include testing liposomal clodronate in xenograft mouse models of other lung cancers...
