Therapeutics: Nuclear paraspeckle assembly transcript 1 (NEAT1)

Patient sample, cell culture and mouse studies suggest inhibiting NEAT1 could help treat breast cancer, squamous cell carcinoma (SCC) of the skin and other cancers. In a human breast cancer cell line, the generic chemotherapy doxycycline increased NEAT1 levels compared with no treatment. Also in the cell line, knockdown of NEAT1 increased sensitivity to veliparib, doxycycline or a p53 inhibitor tool compound compared with normal NEAT1 expression. In a human osteosarcoma cell line treated with the generic chemotherapy bleomycin, NEAT1 knockdown increased markers of DNA damage compared with normal NEAT1 expression. In a mouse model of mutant p53 SCC of the skin, systemic NEAT1 knockout decreased tumor growth and tumor number. Next steps include identifying licensing partners...
