Enchira published in Nature Biotechnology a method for shuffling DNA sequences, RACHITT, which leads to improved recombination frequency (>14 crossovers per gene) and greater exploration of sequence space than existing DNA shuffling technologies. With RACHITT, single-stranded rather than double-stranded fragments are hybridized onto a full-length single-stranded homologous gene, which serves as a scaffold, and resulting fragments are created and separated by size.
