Therapeutics: Adenosine A1 receptor (ADORA1)

Cell culture and rat studies suggest ADORA1 agonists could help treat cisplatin-induced hearing loss. In a cisplatin-treated mouse cochlear hair cell line, pretreatment with an ADORA1 agonist tool compound decreased cisplatin-induced reactive oxygen species (ROS) levels compared with vehicle pretreatment. In a rat model of cisplatin-induced hearing loss, transtympanic injection of the agonist decreased loss of outer cochlear hair cells and increased auditory function compared with vehicle. Next steps could include testing transtympanic injection of other ADORA1 agonists in the model.
