Therapeutics: Mycobacterium tuberculosis cytochrome bc1

In vitro and mouse studies suggest lansoprazole analogs could help treat TB. Screening of a library of FDA-approved drugs in a human cell-based assay of M. tuberculosis infection identified lansoprazole and a lansoprazole sulfide analog as inhibitors of bacterial growth (IC50 values of 1.47 µM and 0.59 µM, respectively). In a mouse model of TB, the analog decreased lung levels of M. tuberculosis compared with vehicle. Next steps could include synthesis of other lansoprazole analogs and testing in models of TB...
