Non-small cell lung cancer (NSCLC)

Notch 1 (NOTCH1)

Cell-based and mouse studies suggest inhibiting NOTCH1 could enhance radiotherapy treatment of NSCLC. In two human NSCLC cell lines, small molecule inhibitors of NOTCH1 plus irradiation decreased proliferation compared with irradiation alone. In a xenograft mouse model of NSCLC, intraperitoneal administration of a NOTCH1 inhibitor in combination with irradiation decreased tumor volume compared with irradiation alone. Next steps could include optimizing NOTCH1 inhibitors.
At least three companies have NOTCH1 inhibitors in preclinical or Phase I development.
