Cell adhesion associated oncogene regulated (CDON; CDO); sonic hedgehog homolog (SHH)

Mouse studies suggest inhibition of CDON could help treat SHH+ tumors. In a mouse model of adenocarcinoma, homozygous deletion of Cdon increased tumor incidence and aggressiveness compared with wild-type Cdon expression. In a mouse xenograft model of lung cancer, small interfering RNA knockdown of Cdon and Shh increased tumor volume compared with knockdown of Shh alone. In a xenograft mouse model of lung cancer, an inhibitor of the SHH-CDON interaction decreased tumor volume compared with saline. Next steps include IND-enabling studies on the identified inhibitor.
