NEDD8 activating enzyme (NAE)

Cell culture studies suggest inhibiting neddylation with NAE inhibitors could help treat inflammation. In a human macrophage-like cell line, the small molecule NAE inhibitor MLN4924 decreased lipopolysaccharide (LPS)-induced expression of proinflammatory cytokines compared with vehicle. Next steps could include testing MLN4924 and other NAE inhibitors in animal models of inflammation.
Takeda Pharmaceutical Co. Ltd.'s Millennium Pharmaceuticals Inc. unit has MLN4924 in Phase I testing to treat various cancers.
