Assay for single-cell DNA methylation analysis

An assay for single-cell DNA methylation analysis could be useful for understanding the heterogeneity of epigenetic modifications in diseased tissue. The assay system combines methylation-sensitive restriction digestion and multiplex, quantitative, real-time PCR in a microfluidic device. The assay showed that early stage mouse embryo cells with a deficiency in the scaffolding protein tripartite motif containing 28 (Trim28) exhibited variable degrees of hypomethylation at all tested genes. Next steps include developing a complete set of assays to monitor methylation status at all genes and adapting the assay for use with human cells.
